چکیده

The synthetic scheme is shown in Supporting Figure S1 and the NMR spectra of new compounds are shown in Supporting Figures S2-S3. In short, rapamycin was acylated with the N-succinimidyl carbonate 1 according to a literature procedure that enables selective modification of the C40 position, yielding the caged rapamycin monomer 2 in 45% yield. This yield is comparable to other reported modifications of rapamycin. The tethered triple bond in 2 was then used in a highly efficient double [3+2] cycloaddition reaction with 1,4-diazidobutane in the presence of CuSO4, sodium ascorbate, and tris[(1-benzyl-1H-1,2,3-triazol-4-yl)methyl]amine (TBTA), delivering the rapamycin dimer dRap in 85% yield.

Light-cleavable rapamycin dimer as an optical trigger for protein dimerization.

Rapamycin-induced protein heterodimerization of FKBP12 and FRB is one of the most commonly employed switches to conditionally control biological processes. We developed an optically activated rapamycin dimer that does not induce FKBP12-FRB dimerization until exposed to light, and applied it to control kinase, protease, and recombinase function.

G-protein Coupled Receptor Dimerization

A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...

Cooperativity and dimerization of recombinant human estrogen receptor hormone-binding domain.

The estrogen receptor dimerizes and exhibits cooperative ligand binding as part of its normal functioning. Interaction of the estrogen receptor with its ligands is mediated by a C-terminal hormone-binding domain (HBD), and residues within the HBD are thought to contribute to dimerization. To examine dimer interactions in the isolated HBD, a human estrogen receptor HBD fragment was expressed in ...

Light regulation of protein dimerization and kinase activity in living cells using photocaged rapamycin and engineered FKBP.

We developed a new system for light-induced protein dimerization in living cells using a photocaged analogue of rapamycin together with an engineered rapamycin binding domain. Using focal adhesion kinase as a target, we demonstrated successful light-mediated regulation of protein interaction and localization in living cells. Modification of this approach enabled light-triggered activation of a ...

Allosteric activation by dimerization of the PknD receptor Ser/Thr protein kinase from Mycobacterium tuberculosis.

To define how extracellular signals activate bacterial receptor Ser/Thr protein kinases, we characterized the regulatory functions of a weak dimer interface identified in the Mycobacterium tuberculosis PknB and PknE receptor kinases. Sequence comparisons revealed that the analogous interface is conserved in PknD orthologs from diverse bacterial species. To analyze the roles of dimerization, we ...
